These neuron-specific filaments are the major constituents of the axonal cytoskeleton . NFs are composed of three major proteins; NF-H, NF-L ａnd NF-M ａnd the ratio of each protein changes during neuronal development. NF-H is the heavy molecular weight protein (200 kDa), NF-M the medium (160 kDa) ａnd NF-L the light protein (68kDa) . NF-L self polymerizes ａnd provides a scaffold for NFs to form via cross-bridging with NF-H or NF-M . NF-L aggregation has been linked to the pathogenesis of neurodegenerative disease, such as degradation of motor neurons .
