產(chǎn)品概述
迪美唑檢測試劑盒酶聯(lián)免疫反應(yīng)測試盒基于競爭性酶聯(lián)反應(yīng)原理,含有迪美唑檢測試劑盒(SEM)的抗體已經(jīng)包被于微孔板上。藥物分析時(shí),樣品同SEM-HRP共同被添加到板孔中。如果樣品中含有SEM,會競爭SEM抗體,抑制SEM-HRP與板上包被的抗體結(jié)合。加入底物后,產(chǎn)物的顏色強(qiáng)弱與樣品中SEM的濃度成反比。

本試劑盒應(yīng)當(dāng)在2-8℃的溫度下儲存,有效期為一年。如果超過3個(gè)月不使用試劑盒,
請將SEM-HRP酶標(biāo)抗原放置-20℃或者冰凍保存。

注意事項(xiàng)
實(shí)驗(yàn)前請閱讀以下文字,以保證獲得*佳實(shí)驗(yàn)效果。
1) 標(biāo)準(zhǔn)品中含有迪美唑檢測試劑盒,請小心使用。
2) 不要使用過期的試劑盒。
3) 不同批次的試劑盒不要混用,抗體和微孔板具有盒與批次的特異性。
4) 盡量保持室溫20-25℃,避免在通風(fēng)口操作防止溫度過低,過熱和/或者蒸發(fā)。同樣的,不要在陽光直射下實(shí)驗(yàn),防止過熱及蒸發(fā)。在孵育期間,如果工作臺溫度過低,應(yīng)該鋪墊若干紙巾或者其他物料。
5) 水質(zhì)量很重要,確保使用蒸餾或者去離子水。
6) 加入樣品或者試劑到空的微孔板時(shí),吸嘴靠于微孔接近底部,并保持接觸。
7) 孵育時(shí)間的計(jì)算越規(guī)范越好,保持添加標(biāo)準(zhǔn)品的一致性,先添加標(biāo)準(zhǔn)品后添加樣品。
8) 從低濃度到高濃度地添加標(biāo)準(zhǔn)品,降低影響標(biāo)準(zhǔn)品曲線質(zhì)量的風(fēng)險(xiǎn)。
9) 將微孔板置于放有干燥劑的密封袋中,冷藏保存。
樣品的準(zhǔn)備【迪美唑檢測試劑盒】
確保樣品的正確保存,一般來說,樣品在2-4只能保存1-2天,如果要保存更長時(shí)間,就要保存在-20冷凍保存,在用之前需要將冷凍的樣品在室溫下或冰箱內(nèi)解凍。
1×樣品提取液
按1:9的比例配制樣品提取濃縮液和雙蒸餾水。
1)飼料(用于檢測添加到飼料中的魚粉、蝦粉或動物組織的SEM)
(1) 取0.5g勻質(zhì)樣品,加入0.5mL1×樣品提取液,3.5mL蒸餾水,0.5mL 1M HCl和20 µL 50 mM 2-硝基苯甲醛,渦旋振蕩1min;
(2) 50°C恒溫培養(yǎng)3小時(shí),在培養(yǎng)期間,每1h振蕩5秒;
(3) 加入5mL 0.1 M K2HPO4,0.4 mL 1 M NaOH,6 mL 乙酸乙酯,轉(zhuǎn)速渦旋震蕩1min;
(4) 室溫下4000g離心10分鐘;
(5) 取3mL乙酸乙酯上清液(含0.25g初始樣品)到另一試管(避免觸及下層水相!如果
移取液中含有水相,再次4000g離心5分鐘,取上層有機(jī)相),在60-70 °C減壓蒸餾或60-70°C氮?dú)獯蹈桑?/span>
(6) 用1 mL 正己烷(或者正庚烷)溶解干燥好的樣品;
(7) 加入1mL1×樣品提取液,轉(zhuǎn)速渦旋振蕩2分鐘;
(8) 室溫下4000g離心10分鐘;
(9) 每孔取100 µL下清液,進(jìn)行樣品測試。
稀釋倍數(shù):4.0
2)魚/蝦/肉(牛肉/雞肉/豬肉):可用于五合一前處理方法
(1) 稱取1g勻漿樣品加入0.5mL1×樣品提取液,3.5mL蒸餾水,0.5mL 1M HCl和20µL50 mM 2-硝基苯甲醛,渦旋震蕩30秒;
(2) 50°C恒溫培養(yǎng)3h,培養(yǎng)期間,每1h振蕩5秒;(說明書后的**標(biāo)記方法可供選擇)
(3) 加入5mL 0.1 M K2HPO4,0.4 mL 1 M NaOH, 6 mL 乙酸乙酯,渦旋震蕩振蕩30s;
(4) 室溫下(20-25℃)4000g離心10分鐘;
(5) 取3mL上清液(包含了0.5g的原始樣品)到另一試管(避免觸及下層水相!如果移取液中含有水相,再次4000g離心5分鐘,取上層有機(jī)相),如果出現(xiàn)乳化現(xiàn)象,上清液不足3mL,請85℃水浴3分鐘。在60 -70°C減壓蒸餾或60-70°C氮?dú)獯蹈桑?/span>
(6) 用1 mL 正己烷(或者正庚烷)溶解干燥好的樣品;(注:肝臟組織富含脂肪、色素等,為防止乳化現(xiàn)象,建議選用4mL正己烷溶解,渦旋震蕩30秒后,再進(jìn)行下一步操作)
(7) 加入1mL1×樣品提取液,劇烈振蕩1分鐘;
(8) 室溫下4000g離心10分鐘;
(9) 每孔取100 µL下清液,進(jìn)行樣品測試。
注意:為了避免過高的背景值,建議用空白樣品做平行,從取3ml乙酸乙酯上清旋轉(zhuǎn)蒸發(fā)開始。
稀釋倍數(shù):2.0
3)雞蛋【迪美唑檢測試劑盒】
(1) 稱取1g雞蛋樣品加入0.5mL1×樣品提取液,3.5mL蒸餾水,0.5mL 1M HCL和20µL 50 mM 2-硝基苯甲醛,劇烈振蕩30秒;
(2) 50°C恒溫培養(yǎng)3小時(shí),在培養(yǎng)期間,每1h振蕩5秒;(說明書后的**標(biāo)記方法可供選擇)
(3) 加入5mL 0.1 M K2HPO4,0.4 mL 1 M NaOH, 6 mL 乙酸乙酯,渦旋震蕩振蕩30s;
(4) 室溫下4000g離心10分鐘;
(5) 取3mL上清液(包含了0.5g的原始樣品) 到另一試管(避免觸及下層水相!如果移取液中含有水相,再次4000g離心5分鐘,取上層有機(jī)相),在60-70℃減壓蒸餾或60-70℃氮?dú)獯蹈桑?/span>
(6) 用1 mL 正己烷(或者正庚烷)溶解干燥好的樣品;
(7) 加入1mL1×樣品抽提工作液體,劇烈振蕩2分鐘;
(8) 室溫下4000g離心10分鐘;
(9) 每孔取100µL下清液,進(jìn)行樣品測試。
稀釋倍數(shù):2.0
注意:為了避免過高的背景值,建議用空白樣品做平行,從取3ml乙酸乙酯上清旋轉(zhuǎn)蒸發(fā)開始。
4)蜂蜜
(1) 稱取1g蜂蜜加入0.5mL1×樣品提取液,3.5mL雙蒸水,0.5mL 1M HCL和20µL 50 mM 2-硝基苯甲醛,劇烈振蕩1分鐘;
(2) 50°C恒溫培養(yǎng)3小時(shí);(說明書后的**標(biāo)記方法可供選擇)
(3) 加入5mL 0.1 M K2HPO4,0.4 mL 1 M NaOH , 6 mL 乙酸乙酯,轉(zhuǎn)速振蕩1min;
(4) 室溫下4000g離心10分鐘;
(5) 取3mL上清液(包含了0.5g的原始樣品) 到另一試管(避免觸及下層水相!如果移取液中含有水相,再次4000g離心5分鐘,取上層有機(jī)相),在60 -70°C減壓蒸餾或60-70°C氮?dú)獯蹈桑?/span>
(6) 用1 mL 正己烷(或者正庚烷)溶解干燥好的樣品;

5)牛奶
(1) 對于含脂牛奶,取待測牛奶樣品3ml室溫4000g,5min。除去上層脂肪層。(對于脫脂奶粉,次步略)
(2) 取1ml樣品,加入0.5mL1×樣品提取液,3.5mL雙蒸水,0.5mL 1M HCL和40µL 50 mM 2-硝基苯甲醛,劇烈振蕩1分鐘;
(3) 50°C恒溫培養(yǎng)3小時(shí);(說明書后的**標(biāo)記方法可供選擇)
(4) 加入5mL 0.1 M K2HPO4,0.4 mL 1 M NaOH,6 mL 乙酸乙酯,轉(zhuǎn)速振蕩1min;室溫下4000g離心10分鐘;
(5) 取3mL上清液(包含了0.5g的原始樣品) 到另一試管(避免觸及下層水相!如果移取液中含有水相,再次4000g離心5分鐘,取上層有機(jī)相),在60 -70°C減壓蒸餾或60-70°C氮?dú)獯蹈桑?/span>

(1) 稱取1g血清加入0.5mL1×樣品提取液,3.5mL雙蒸水,0.5mL 1M HCL和20µL 50 mM 2-硝基苯甲醛,劇烈振蕩1分鐘;
(2) 50°C恒溫培養(yǎng)3小時(shí);(說明書后的**標(biāo)記方法可供選擇)
(3) 加入5mL 0.1 M K2HPO4,0.4 mL 1 M NaOH , 6 mL 乙酸乙酯,轉(zhuǎn)速振蕩1min;
(4) 室溫下4000g離心10分鐘;
(5) 取3mL上清液(包含了0.5g的原始樣品) 到另一試管(避免觸及下層水相!如果移

7)尿樣
(1) 取3ml尿樣,4000g離心5min。
(2) 離心后稱取1ml,加入0.5mL1×樣品提取液,3.5mL雙蒸水,0.5mL 1M HCL和40µL 50 mM 2-硝基苯甲醛,劇烈振蕩1分鐘;
(3) 50°C恒溫培養(yǎng)3小時(shí);(說明書后的**標(biāo)記方法可供選擇)
(4) 加入5mL 0.1 M K2HPO4,0.4 mL 1 M NaOH , 6 mL 乙酸乙酯,轉(zhuǎn)速振蕩1min;
(5) 室溫下4000g離心10分鐘;
(6) 取3mL上清液(包含了0.5g的原始樣品) 到另一試管(避免觸及下層水相!如果移取液中含有水相,再次4000g離心5分鐘,取上層有機(jī)相),在60 -70°C減壓蒸餾或60-70°C氮?dú)獯蹈桑?/span>

檢測步驟
試劑的準(zhǔn)備
注意:試劑在使用之前要在室溫下解凍1-2小時(shí),確定在使用前閱讀過注意事項(xiàng)。準(zhǔn)備適量的檢測用試劑,所有的試劑搖勻后使用。準(zhǔn)備足夠所有小管所需的用量,不能將試劑倒回原來的瓶子中,處理試劑時(shí)建議用廢棄的瓶子以降低污染的風(fēng)險(xiǎn)。

(3) 在每孔中加入50mL 1×SEM-HRP,輕敲微孔板邊緣混勻1分鐘;
(4) 室溫(22.5±2.5)°C避光孵育30分鐘;
(5) 洗板3次,每次應(yīng)該加入250mL 1×洗液,*后一次使板盡量甩干并在吸水紙上吸干;
(6) 每孔加入100mL的TMB底物;
(7) 在室溫(22.5±2.5)°C避光孵育20分鐘后,每孔加入100mL的終止液終止反應(yīng);
(8) 在450nm或450/650nm下讀OD值 (讀數(shù)前,用吸水紙將板底部的指痕和水分擦去,避免影響讀數(shù))。
10.結(jié)果計(jì)算【迪美唑檢測試劑盒】
(1) 分別計(jì)算標(biāo)準(zhǔn)、質(zhì)控和樣品的平均吸光度值和相對吸光度值。
相對吸光度值 (%) = (標(biāo)準(zhǔn)或樣品吸光度值/零標(biāo)準(zhǔn)吸光度值) ´ 100
(2) 以相對吸光度值為縱坐標(biāo),標(biāo)準(zhǔn)濃度為橫坐標(biāo)建立標(biāo)準(zhǔn)曲線。
(3) 從標(biāo)準(zhǔn)曲線上讀取質(zhì)控和樣品的濃度值。
(4) 樣品檢測下限計(jì)算方法如下:
樣品檢測下限 = (0.025ng/g) ´ (稀釋倍數(shù))
樣品定量下限 = (0.1ng/g) ´ (稀釋倍數(shù))
例如,肉類樣品的稀釋倍數(shù)是2.0,那么肉類樣品的檢測下限是0.05ppb,定量檢測下限是0.2ppb。
