ELISA檢測(cè)試劑盒應(yīng)用定性?shī)A心免疫檢測(cè)技術(shù),用合成的HEV多肽抗原包被微孔板板條,這些多肽是中國(guó)型HEV毒株核心氨基酸序列中抗原性很強(qiáng)的肽段,分別來(lái)自于該毒株的開(kāi)放閱讀框2和開(kāi)放閱讀框3.將樣品或標(biāo)準(zhǔn)品加入孔中并孵育,如果其中存在HEVIgM抗體,這些抗體就會(huì)與HEV的多肽抗原結(jié)合,并固定在上面,洗板除去其它非特異性抗體和樣品中的其它成份.然后加入羊抗人IgM-HRP（辣根過(guò)氧化物酶）酶結(jié)合物,經(jīng)第二次孵育后,酶結(jié)合物就會(huì)與次孵育結(jié)合上的HEVIgM抗體相結(jié)合,洗板除去未結(jié)合的酶結(jié)合物,加入TMB底物溶液,在第三次孵育時(shí)會(huì)發(fā)生酶-底物反應(yīng),只有那些含有HEVIgM抗體和酶結(jié)合物所形成的復(fù)合物的孔才會(huì)發(fā)生顏色變化,加入硫酸溶液終止酶和底物間的反應(yīng),并在波長(zhǎng):450nm處測(cè)量O.D.值,按照本HEVIgM抗體elisa試劑盒的測(cè)試標(biāo)準(zhǔn),O.D.值大于或等于Cut-Off值的樣品被認(rèn)為是初試陽(yáng)性.
