二、實驗材料與試劑配制：

1. 儀器與材料：酶標(biāo)儀（使用前預(yù)熱30分鐘），微量加液器、吸頭、蒸餾水或去離子水，濾紙。

2. 緩沖液使用：加5ul  的緩沖液于50ul  的樣品中，如果樣品量不夠或者不確定，緩沖液和樣品的混合比例不要小于1:10即可混勻，靜置1小時備用（如果標(biāo)本是血清或者血漿，此步驟忽略）。

3. 洗液的配制：按1:100的比例配制洗液備用。

1.檢查試劑盒標(biāo)簽上的有效期。如果超過有效期，請勿使用。

2. 按說明書確定所有試劑齊全（數(shù)量、體積）

3. 標(biāo)本制備要規(guī)范，每份標(biāo)本體積要按2-3個復(fù)孔以上的量制備（貯存），盡量分裝做備份。

4.短期內(nèi)無法實驗者，注意低溫保存

5.準(zhǔn)備好所有實驗額外所需物品，（比如移液器，試管，清洗器，酶標(biāo)儀）

6. 按說明書將所用試劑平衡至室溫，根據(jù)檢測標(biāo)本數(shù)量確定所需試劑的量

8.檢查不穩(wěn)定或者變質(zhì)的試劑溶液（比如沉淀或變色）。有些試劑，比如標(biāo)準(zhǔn)品稀釋液或者檢測稀釋液，可能在設(shè)計時就含有沉淀物。所有試劑在滴入酶標(biāo)板之前，必需充分混勻。而由于沉淀物的特性，在加入酶標(biāo)板之前，必需不停攪拌。

9.保證充分的孵育時間和溫度。

10.切勿使用不同生產(chǎn)批號的試劑取代現(xiàn)有試劑或混合現(xiàn)有試劑。

11.在混合或溶解蛋白溶液時，避免泡沫產(chǎn)生。

12.在實驗開始之前，安排好實驗流程。在實驗開始之前，清潔工作臺。

1.在收集標(biāo)本前都必須有一個完整的計劃，必須清楚要檢測的成份是否足夠穩(wěn)定。我們提倡新鮮標(biāo)本盡早檢測，對收集后當(dāng)天就進行檢測的標(biāo)本，及時儲存在4℃?zhèn)溆�，如有特殊原因需要周期收集�?biāo)本，請造模取材后，將標(biāo)本及時分裝后放在-20℃或-70℃條件下保存。因冰室與室溫存在一定溫差，蛋白極易降解，直接影響實驗質(zhì)量，所以避免反復(fù)凍融。部分標(biāo)本也可加入適當(dāng)防腐劑，激素類標(biāo)本需添加抑肽酶。 

2.試劑盒代測標(biāo)本的客戶取材前須向我司銷售人員索要說明書，具體操作注意事項請與我司技術(shù)人員溝通。

3.液體類標(biāo)本：標(biāo)本必須為液體，不含沉淀。包括血清、血漿、尿液、胸腹水、腦脊液、細胞培養(yǎng)上清、組織勻漿等。

For defect DML, after customer calling QIAGEN technical service hotline before 12 am:

- A special package containing a replacement unit shall be sent to the customer within 2 working days.

- Customer shall send the defect unit to QIAGEN Regional Repair Center following the instructions attached with the packaging.   

For DML owned by customer, QIAGEN Regional Repair Center will send back the repaired original unit to customers within 7-10 working days.

PC, Monitor, Heater, Shaker, Washer, Printer ａnd Votexer:

- A new unit shall be sent to customer within  7-10 working days at no extra cost

- Customer shall send the defect unit to QIAGEN for disposal

- On-site installation of PC ａnd application software is not covered by the agreement

The Preventive Maintenance (PM) service ensures the correct operation of the equipment. During the PM, all the instrument modules are inspected by a qualified QIAGEN service technician.
