二、實驗材料與試劑配制：

1. 儀器與材料：酶標儀（使用前預熱30分鐘），微量加液器、吸頭、蒸餾水或去離子水，濾紙。

2. 緩沖液使用：加5ul  的緩沖液于50ul  的樣品中，如果樣品量不夠或者不確定，緩沖液和樣品的混合比例不要小于1:10即可混勻，靜置1小時備用（如果標本是血清或者血漿，此步驟忽略）。

3. 洗液的配制：按1:100的比例配制洗液備用。

1.檢查試劑盒標簽上的有效期。如果超過有效期，請勿使用。

2. 按說明書確定所有試劑齊全（數(shù)量、體積）

3. 標本制備要規(guī)范，每份標本體積要按2-3個復孔以上的量制備（貯存），盡量分裝做備份。

4.短期內無法實驗者，注意低溫保存

5.準備好所有實驗額外所需物品，（比如移液器，試管，清洗器，酶標儀）

6. 按說明書將所用試劑平衡至室溫，根據(jù)檢測標本數(shù)量確定所需試劑的量

8.檢查不穩(wěn)定或者變質的試劑溶液（比如沉淀或變色）。有些試劑，比如標準品稀釋液或者檢測稀釋液，可能在設計時就含有沉淀物。所有試劑在滴入酶標板之前，必需充分混勻。而由于沉淀物的特性，在加入酶標板之前，必需不停攪拌。

9.保證充分的孵育時間和溫度。

10.切勿使用不同生產(chǎn)批號的試劑取代現(xiàn)有試劑或混合現(xiàn)有試劑。

11.在混合或溶解蛋白溶液時，避免泡沫產(chǎn)生。

12.在實驗開始之前，安排好實驗流程。在實驗開始之前，清潔工作臺。

1.在收集標本前都必須有一個完整的計劃，必須清楚要檢測的成份是否足夠穩(wěn)定。我們提倡新鮮標本盡早檢測，對收集后當天就進行檢測的標本，及時儲存在4℃?zhèn)溆�，如有特殊原因需要周期收集標本，請造模取材后，將標本及時分裝后放在-20℃或-70℃條件下保存。因冰室與室溫存在一定溫差，蛋白極易降解，直接影響實驗質量，所以避免反復凍融。部分標本也可加入適當防腐劑，激素類標本需添加抑肽酶。 

2.試劑盒代測標本的客戶取材前須向我司銷售人員索要說明書，具體操作注意事項請與我司技術人員溝通。

3.液體類標本：標本必須為液體，不含沉淀。包括血清、血漿、尿液、胸腹水、腦脊液、細胞培養(yǎng)上清、組織勻漿等。
