INTENDED USE：

The Human CP ELISA is a highly sensitive two-site enzyme-linked immunoassay (ELISA) for the quantitative

determination of CP in human biological samples. For research use only. Not for use in diagnostic procedures.

INTRODUCTION：

Ceruloplasmin is a multifunctional protein involved in copper transport ａnd is also an important serum antioxidant. During inflammation, traumatic injury, or myocardial infarction, the serum levels of ceruloplasmin, as an acute phase reactant,rise 2-3 fold.

PRINCIPLE：

The principle of the double antibody sandwich ELISA is represented in Figure 1. In this assay the CP present in samples reacts with the anti-CP antibodies which have been adsorbed to the surface of polystyrene microtiter wells. After the removal of unbound proteins by washing, the Detection Antibody, biotin conjugated anti-CP is added ａnd complexes are formed. Following a wash step, the horseradish peroxidase (HRP) conjugated Streptavidin is added ａnd complexes are formed. After another washing step, the complexes are assayed by the addition of a chromogenic substrate, 3,3’,5,5’- tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of CP in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of CP in the test sample. The quantity of CP in the sample can be interpolated from the calibration curve constructed from the calibrators, ａnd corrected for sample dilution.   

近年來(lái)研究結(jié)果認(rèn)為銅藍(lán)蛋白起著抗氧化劑的作用。在血循環(huán)中銅藍(lán)蛋白的抗氧化活力可以防止組織中脂質(zhì)過(guò)氧化物和自由基的生成，特別在炎癥時(shí)具有重要意義。銅藍(lán)蛋白也屬于一種急性時(shí)相反應(yīng)蛋白。血漿銅藍(lán)蛋白在感染、創(chuàng)傷和腫瘤時(shí)增加。其特殊的作用在于協(xié)助Wilson病的診斷，即患者血漿銅藍(lán)蛋白含量明顯下降，而伴有血漿可透析的銅含量增加。大部分患者可有肝功能損害并伴有神經(jīng)系統(tǒng)的癥狀，如不及時(shí)治療，此病是進(jìn)行性和致命的。血漿銅藍(lán)蛋白在營(yíng)養(yǎng)不良、嚴(yán)重肝病及腎病綜合征時(shí)亦往往下降。婦女妊娠期、口服避孕藥時(shí)其含量有明顯增加。

艾美捷KT-1876人銅藍(lán)蛋白ELISA試劑盒是一種高度靈敏的雙位點(diǎn)酶聯(lián)免疫分析（ELISA）人類(lèi)生物樣品中CP的測(cè)定。僅供研究使用。不用于診斷程序。

人銅藍(lán)蛋白ELISA試劑盒組成：

1.濃縮稀釋劑

一瓶含有50mL 5X濃縮稀釋液流動(dòng)緩沖液。

2.濃縮洗滌液

一瓶含有50mL 20X濃縮洗滌液。

3.檢測(cè)抗體100X

一個(gè)小瓶含有150µL親和純化的抗人CP抗體，該抗體在穩(wěn)定緩沖液中與生物素綴合。

4.HRP鏈親和素100X

一個(gè)小瓶，在穩(wěn)定緩沖液中含有150µL HRP結(jié)合的鏈霉親和素。

5.TMB基板溶液

一瓶含有12mL TMB和過(guò)氧化氫的檸檬酸緩沖液（pH 3.3）。

6.S Solution

一瓶含12 mL 0.3 M硫酸。

警告：避免接觸皮膚。

7.微量滴定板

井支架框架中的十二個(gè)可拆卸的八（8）個(gè)井微孔條。每個(gè)孔用親和純化的抗人CP包被。

8.人體CP校準(zhǔn)器

一個(gè)裝有凍干人CP校準(zhǔn)品的小瓶。

人銅藍(lán)蛋白ELISA試劑盒標(biāo)本采集和處理：

應(yīng)通過(guò)靜脈穿刺收集血液，并在凝塊形成后通過(guò)離心將血清與細(xì)胞分離。對(duì)于血漿樣本，應(yīng)將血液收集到裝有抗凝劑的容器中，然后離心。應(yīng)注意為了盡量減少溶血，過(guò)量的溶血會(huì)影響您的結(jié)果。立即測(cè)定或等分并儲(chǔ)存-20°C下的樣品。避免重復(fù)凍融循環(huán)。

對(duì)于任何可能含有病原體的樣本，必須小心防止與開(kāi)放性傷口接觸。無(wú)添加劑或防腐劑是保持標(biāo)本完整性所必需的。避免疊-氮-化物污染。
