這種靶向毒素(分子量31.8kDa)靶向GRP受體陽(yáng)性細(xì)胞。Bombesin SAP是一家蛙皮素和核糖體失活蛋白saporin(Cat.#PR-01)的化學(xué)綴合物。
Advanced Targeting Systems/ATSbio艾美捷蛙皮素SAP使用和儲(chǔ)存:
Bombesin SAP特異性靶向GRP受體陽(yáng)性細(xì)胞。在布料工作稀釋度必須由終用戶確定。如果這是一個(gè)新批次,您必須評(píng)估在開始完整的實(shí)驗(yàn)方案之進(jìn)行適當(dāng)?shù)墓ぷ飨♂尅J褂幂p輕旋轉(zhuǎn)材料;5-10的縮微膠片就足夠了。將材料儲(chǔ)存在未稀釋的等分試樣在-20°C下保存1-2個(gè)月。為了長(zhǎng)期儲(chǔ)存,將材料儲(chǔ)存在–80°C。布料應(yīng)等分至方便的體積和數(shù)量,以避免重復(fù)冷凍和解凍損害蛋白質(zhì)含量。在這些條件下,該材料具有非常穩(wěn)定的保質(zhì)期。解凍應(yīng)該是在室溫或冰上進(jìn)行。解凍的溶液在使用應(yīng)保持在冰上。
請(qǐng)勿將還原劑(如二硫蘇糖醇、β-巰基乙醇或抗壞血酸)用于此材料。它將使毒素失活。這種物質(zhì)是一種強(qiáng)的細(xì)胞毒素。應(yīng)由經(jīng)驗(yàn)豐富的人員進(jìn)行搬運(yùn)。手套和處理本產(chǎn)品時(shí)需要佩戴安全眼鏡。處置時(shí)必須小心;高壓滅菌或暴露至0.2M氫氧化鈉將使材料失活。所有與此材料接觸的實(shí)驗(yàn)室用品應(yīng)同樣處理。
圖:T47D細(xì)胞,一種人上皮細(xì)胞系以2500個(gè)細(xì)胞/孔/90ul鋪板孵育過(guò)夜。第二天,Saporin(PR-01)和Bombesin SAP(IT-40)以10μl的體積添加劑量范圍為1uM-100fM和100nM-10fM,分別地平板在37°C下孵育72小時(shí)。將XTT/PMS混合物以50μl的體積添加到每個(gè)孔中在450nm下讀取板,并用Prism進(jìn)行數(shù)據(jù)分析軟件(GraphPad,圣地亞哥)。

Objective: To investigate the molecular mechanisms of how IL-31 is produced in helper T cells upon stimulation and transmits the itch sensation to the brain.
Summary: This review highlights recent findings that show the functional significance of endothelial PAS domain 1 (EPAS1) and neurokinin B (NKB) in the IL-31-induced itch sensation.
Usage: Neurons expressing the Nppb receptor were specifically ablated by intrathecal injection of Nppb-SAP. Treatment with Bombesin-SAP reduced IL-31-induced scratching.
