Camptothecin is a potent cytotoxic pentacyclic alkaloid known for its potent apoptosis inducing properties. Camptothecin ａnd its analogs have been demonstrated to inhibit oisomerase I (o I), causing cells in S phase to enter into apoptosis. Camptothecin binds to ａnd stabilizes the o I – DNA backbone complex (covalent binary complex). This stabilized complex diminishes the rate of o I release from the broken strand, thus slowing the religation ａnd subsequent DNA synthesis process. Use camptothecin to create positive controls in apoptosis detection experiments.

喜樹堿是一種植物抗癌藥物，從中國(guó)中南、西南分布的喜樹中提取得到。1976年中國(guó)化學(xué)家高怡生等合成消旋喜樹堿成功。喜樹堿對(duì)腸胃道和頭頸部癌等有較好的近期療效，但對(duì)少數(shù)病人有尿血的副作用。10-羥基喜樹堿的抗癌活性超過(guò)喜樹堿，對(duì)肝癌和頭頸部癌也有明顯療效 ，而且副作用較少。

艾美捷喜樹堿是一種具有抗腫瘤特性的細(xì)胞毒性植物生物堿，是一種原型DNA拓?fù)洚悩?gòu)酶I抑制劑。在凋亡檢測(cè)實(shí)驗(yàn)中使用喜樹堿創(chuàng)建陽(yáng)性對(duì)照。

艾美捷喜樹堿(Camptothecin)基本操作：

1、將喜樹堿粉末溶解在組織培養(yǎng)DMSO中，以獲得2mg/mL喜樹堿儲(chǔ)備濃度。

2、制備50–100µL的DMSO增溶喜樹堿等分試樣，并將其冷凍保存在<-20°C。一瓶冷凍的喜樹堿可在丟棄再解凍2次。應(yīng)標(biāo)記已解凍1X的小瓶，以表明這一點(diǎn)，以便它們?cè)趤G棄只經(jīng)過(guò)一次解凍。

3、喜樹堿濃度范圍為2–4µg/mL的刺突細(xì)胞培養(yǎng)物，產(chǎn)生6–12µM的細(xì)胞培養(yǎng)物喜樹堿濃度。該喜樹堿濃度范圍適用于誘導(dǎo)Jurkat或HL 60細(xì)胞懸浮液，其濃度范圍通常為1 x 105–1 x 106細(xì)胞/mL。在此濃度范圍內(nèi)，培養(yǎng)4小時(shí)（37°C）后成功誘導(dǎo)凋亡。

4、對(duì)特定細(xì)胞系進(jìn)行時(shí)間歷程研究，以確定合適的喜樹堿濃度和暴露時(shí)間，從而為實(shí)驗(yàn)實(shí)現(xiàn)良好的凋亡誘導(dǎo)水平。
