FAM FLICA Caspase-1探針允許研究人員評(píng)估Caspase-1的激活。FLICA試劑FAM-YVAD-FMK進(jìn)入每個(gè)細(xì)胞,并不可逆地與活化的caspase-1結(jié)合。因?yàn)?span style="font-family: Calibri;">FAM-YVAD-FMK FLICA試劑與活性酶共價(jià)偶聯(lián),所以它保留在細(xì)胞內(nèi),而任何未結(jié)合的FAM-YVAD-FMK-FLICA試劑擴(kuò)散出細(xì)胞并被沖走。剩余的綠色熒光信號(hào)是添加試劑時(shí)細(xì)胞中存在的活性caspase-1酶活性的直接測(cè)量。含有結(jié)合的FLICA的細(xì)胞可通過(guò)熒光平板讀取器、熒光顯微鏡或流式細(xì)胞儀進(jìn)行分析。用FLICA試劑標(biāo)記的細(xì)胞可立即讀取或使用試劑盒中的固定劑保存16小時(shí)。還可以用碘化丙啶或Hoechst 33342分析未固定的樣品,以分別檢測(cè)核形態(tài)的壞死或變化。
