圖2：使用1000ng/mL的B細(xì)胞活化試驗(yàn)對(duì)連續(xù)7個(gè)生產(chǎn)批次的MEGACD40L®進(jìn)行測(cè)試。圖1：通過(guò)MEGACD40L®（ALX-522-110）激活B細(xì)胞淋巴細(xì)胞。方法：將外周血單核細(xì)胞（PBMC）在37°C、5%CO2下在48孔板（1 x 106個(gè)細(xì)胞/孔）中孵育48小時(shí)。每個(gè)孔含有200µl無(wú)血清測(cè)試培養(yǎng)基和連續(xù)稀釋的MEGACD40L®。處理后，清洗細(xì)胞，用小鼠抗人CD19–PE和小鼠抗人CD 86–APC雙重染色，并通過(guò)流式細(xì)胞術(shù)進(jìn)行分析。數(shù)據(jù)以每種MEGACD40L®濃度下共染色為CD86陽(yáng)性的CD19陽(yáng)性B細(xì)胞的百分比表示。
