原理 人血清蛋白酶聯(lián)免疫吸附試驗(yàn)是一種基于三明治原理的即用型固相酶聯(lián)免疫吸附試驗(yàn)，工作時(shí)間為3個(gè)半小時(shí)。由12個(gè)一次性8孔條組成的板塊的有效形式允許自由選擇檢測的批量大小。樣品和標(biāo)準(zhǔn)品在涂有識別人MPO的抗體的微孔中孵化。生物素化的示蹤劑抗體將與捕獲的人MPO結(jié)合。鏈霉親和素-過氧化物酶結(jié)合物將與生物素化示蹤抗體結(jié)合。鏈霉蛋白-過氧化物酶結(jié)合物將與底物四甲基聯(lián)苯胺（TMB）反應(yīng)。酶的反應(yīng)通過加入草酸停止。用分光光度計(jì)測量450納米處的吸光度。通過繪制吸光度（線性）與人類MPO標(biāo)準(zhǔn)品的相應(yīng)濃度（對數(shù)）的關(guān)系，得到標(biāo)準(zhǔn)曲線。與標(biāo)準(zhǔn)品同時(shí)進(jìn)行的樣品的人類MPO濃度可由標(biāo)準(zhǔn)曲線確定。
