試劑盒是固相夾心法酶聯(lián)免疫吸附實(shí)驗(yàn)（ELISA）.已知濃度的標(biāo)準(zhǔn)品、未知濃度的樣品加入微孔酶標(biāo)板內(nèi)進(jìn)行檢測.先將生物素標(biāo)記的抗體同時(shí)溫育.洗滌后,加入親和素標(biāo)記過的HRP.再經(jīng)過溫育和洗滌,去除未結(jié)合的酶結(jié)合物,然后加入底物A、B,和酶結(jié)合物同時(shí)作用.產(chǎn)生顏色.顏色的深淺和樣品中的濃度呈比例關(guān)系.

HBsAg試劑特點(diǎn)（檢測模式：臨床抗體夾心法）：包被抗體為山羊多抗.多抗具有高親和性和對抗原各種表位的反應(yīng)性.酶表抗體為與HBsAg有不同結(jié)合位點(diǎn)的鼠復(fù)合單位,可測得HBsAg變異樣品,提高檢測的特異性（99.98%）提高檢測的靈敏度,應(yīng)用ParlEhrich學(xué)說（PEI）HBsAg標(biāo)準(zhǔn)品,對ad標(biāo)準(zhǔn)品的靈敏度為0.05ng/ml對ay標(biāo)準(zhǔn)品靈敏度為0.025ng/ml

ELISA檢測試劑盒應(yīng)用定性夾心免疫檢測技術(shù),用合成的HEV多肽抗原包被微孔板板條,這些多肽是中國型HEV毒株核心氨基酸序列中抗原性很強(qiáng)的肽段,分別來自于該毒株的開放閱讀框2和開放閱讀框3.將樣品或標(biāo)準(zhǔn)品加入孔中并孵育,如果其中存在HEVIgM抗體,這些抗體就會與HEV的多肽抗原結(jié)合,并固定在上面,洗板除去其它非特異性抗體和樣品中的其它成份.然后加入羊抗人IgM-HRP（辣根過氧化物酶）酶結(jié)合物,經(jīng)第二次孵育后,酶結(jié)合物就會與次孵育結(jié)合上的HEVIgM抗體相結(jié)合,洗板除去未結(jié)合的酶結(jié)合物,加入TMB底物溶液,在第三次孵育時(shí)會發(fā)生酶-底物反應(yīng),只有那些含有HEVIgM抗體和酶結(jié)合物所形成的復(fù)合物的孔才會發(fā)生顏色變化,加入硫酸溶液終止酶和底物間的反應(yīng),并在波長:450nm處測量O.D.值,按照本HEVIgM抗體elisa試劑盒的測試標(biāo)準(zhǔn),O.D.值大于或等于Cut-Off值的樣品被認(rèn)為是初試陽性.
