注意事項(xiàng)：1.重復(fù)冷凍/融化循環(huán)可使抗體變性，導(dǎo)致其形成使抗體結(jié)合能力降低的聚集物。保存于-20℃對(duì)于大多數(shù)抗體是適當(dāng)?shù)�；保存�?80℃無(wú)明顯優(yōu)勢(shì)。冰箱不能為無(wú)霜型。這些冷凍和融化之間的循環(huán)（以減少結(jié)霜）恰恰是應(yīng)該避免的。2.抗體試劑瓶應(yīng)置于具有*小溫度波動(dòng)的冰箱區(qū)域，例如朝冰箱后部放置而不是置于門(mén)架上。有些研究人員加入冷凍保護(hù)劑甘油達(dá)到50%的終濃度以防止冷凍/融化損傷；甘油可將凝固點(diǎn)降低至低于-20℃。雖然這對(duì)于很多抗體是可接受的，但是只有一小部分我們提供的抗體測(cè)試了在該保存條件下的穩(wěn)定性，并且我們的保證僅使用于按數(shù)據(jù)表上的方式保存的抗體。不建議將包含甘油的溶液保存于-80℃，因?yàn)樵摐囟鹊陀诟视偷哪厅c(diǎn)。請(qǐng)注意甘油可被細(xì)菌污染，如果加入甘油或任何冷凍保護(hù)劑，則應(yīng)當(dāng)小心操作，以得到無(wú)菌制備物。3.應(yīng)避免抗體在稀釋至工作濃度后在4℃下保存超過(guò)一天。蛋白質(zhì)在高濃度下保存時(shí)一般不易降解，理想的濃度為1mg/ml或更高。這是在抗體溶液中加入蛋白質(zhì)例如BSA作為穩(wěn)定劑的理論基礎(chǔ)。加入的蛋白質(zhì)也有助于減少抗體因結(jié)合于容器壁上而造成的損失。對(duì)于打算進(jìn)行偶聯(lián)的抗體而言，由于穩(wěn)定蛋白質(zhì)會(huì)與抗體競(jìng)爭(zhēng)，降低了偶聯(lián)的效率，所以不應(yīng)加入穩(wěn)定蛋白質(zhì)。溫馨提示：在實(shí)驗(yàn)后，請(qǐng)注意回收稀釋的抗體�；厥盏目贵w在進(jìn)行Western實(shí)驗(yàn)時(shí)至少可以重復(fù)使用10次。稀釋后的抗體，包括已經(jīng)使用過(guò)的稀釋抗體，4℃保存。回收后重復(fù)使用的抗體，使用方法同新鮮稀釋的抗體。如果在重復(fù)使用過(guò)程中發(fā)現(xiàn)抗體出現(xiàn)輕微混濁現(xiàn)象，可以10000g離心1-3分鐘，取上清用于后續(xù)檢測(cè)。如果回收的抗體出現(xiàn)明顯的絮狀物或長(zhǎng)霉長(zhǎng)菌等情況，則可以考慮廢棄該抗體。
