描述： Alzheimer＇s disease is a common neurodegenerative, progressive, ａnd fatal disorder. Generation ａnd deposition of amyloid beta (Aβ) peptides associate with its pathogenesis ａnd small soluble Aβ oligomers show the most pronounced neurotoxic effects ａnd correlate with disease initiation ａnd progression.
Beta amyloid 1-42 is a 42-amino acid peptide which plays a key role in the pathogenesis of Alzheimer, even slightly increased amounts of this Aβ(1–42) might be sufficient to cause Alzheimer＇s disease (AD).

Beta amyloid proteins 聚集指南（以下方案僅供參考，請根據(jù)您的具體實(shí)驗需求進(jìn)行調(diào)整）:
1. 將Aβ肽(1-42)粉末溶于冷的六氟-2-丙醇（HFIP）。肽溶液在室溫下孵化至少1小時，以建立單體化和隨機(jī)化的結(jié)構(gòu)。(Solid Aβ peptide was dissolved in cold hexafluoro-2-propanol (HFIP). The peptide was incubated at room temperature for at least 1h to establish monomerization ａnd randomization of structure.)
2. 蒸發(fā)掉HFIP，將得到的多肽以薄膜形式保存在-20或-80℃。(The HFIP was removed by evaporation, ａnd the resulting peptide was stored as a film at -20 or -80 ℃.)
3. 將得到的薄膜以5 mM的濃度溶解于無水DMSO中，然后用渦旋方式稀釋到適當(dāng)?shù)臐舛群途彌_液（不含血清和酚紅的培養(yǎng)基）中。(The resulting film was dissolved in anhydrous DMSO at 5 mM ａnd then diluted into the appropriate concentration ａnd buffer (serum- ａnd phenol red-free culture medium) with vortexing.)
4. 接下來，該溶液在4-8℃下老化48h。然后在4-8 ℃下以14000g離心10分鐘；可溶性低聚物在上清液中。將上清液稀釋10-200倍用于后續(xù)實(shí)驗。(Next, the solution was age 48h at 4-8 ℃. The sample was then centrifuged at 14000g for 10 min at 4-8 ℃; the soluble oligomers were in the supernatant. The supernatant was diluted 10-200-fold for experiments.)
