艾美捷重組蛋白酶K是一種不含動物源性物質的(animal origin-free, AOF)的蛋白酶K,通過畢赤酵母(Pichia pastoris)發(fā)酵并純化獲得。

盡管鈣離子不影響酶的活性，但當濃度為1-5μmol時，它們確實有助于酶的穩(wěn)定性。蛋白酶K的一個有趣的特征是，它在十二烷基硫酸鈉（SDS）或尿素存在下保持活性。（0.5-1%SDS和1-4M尿素）。將反應溫度從37℃提高到50℃-60℃可以將活性提高幾倍。蛋白酶K的一個特殊特征是其消化天然蛋白質的能力，從而使酶如DNase和RNase失活，而無需借助變性過程。

艾美捷重組蛋白酶K，無動物源/AF穩(wěn)定性/儲存：凍干粉末在2-8°C下穩(wěn)定一年。50mM Tris-HCl，pH 8.0和1mM CaCl2的溶液在2-8°C下穩(wěn)定數(shù)月。儲存于2-8°C。

艾美捷重組蛋白酶K，無動物源/AF單位定義：在37°C，pH 7.5，使用尿素變性血紅蛋白作為底物，One Unit釋放1微摩爾Folin陽性氨基酸（以酪氨酸計）。
