C1q蛋白家族由眾多含C1q結(jié)構(gòu)域的蛋白組成, 從細(xì)菌到高等哺乳動物中都有分布。這類蛋白由一條信號肽、膠原樣區(qū)(Collage-like region, CLR)和C1q球狀結(jié)構(gòu)域(Globular C1q domain, gC1q)組成。C1q蛋白家族根據(jù)其結(jié)構(gòu)特點(diǎn), 可分為三大類分子：C1q、C1q-like和ghC1q。C1q是補(bǔ)體經(jīng)典途徑的起始分子, 能夠識別免疫復(fù)合物, 啟動補(bǔ)體系統(tǒng)經(jīng)典途徑; 此外, 作為一種模式識別受體分子(Pattern recognition receptor, PRR), 它可以結(jié)合種類繁多的配體。

艾美捷C1q天然蛋白的應(yīng)用：

C1q的生物活性定義為產(chǎn)生50%致敏紅細(xì)胞（EA）裂解所需的C1q量。對于功能性研究，體外稀釋必須在用戶的實(shí)驗(yàn)環(huán)境中進(jìn)行優(yōu)化。

Formulation 1 ml of approximately 1 mg/ml purified natural human C1q isolated from healthy blood donors, in 10 mM HEPES, 300 mM NaCl, pH 7.2. The blood donors have been tested ａnd found negative for various viruses (see table: Human blood test results). The functional activity is indicated on the label.

Use For functional studies, in vitro dilutions have to be optimized in user＇s experimental setting.

Application Functional studies

Storage ａnd stability Product should be stored at -70 °C. Repeated freeze ａnd thaw cycles will cause loss of activity. The exact expiry date is indicated on the label. Use C1q protein within 24 hours after thawing ａnd keep on ice. Remainder amounts should be aliquoted ａnd immediately re-freezed for future use. Aliquots should never be thawed more than once. Under recommended storage conditions, product is stable for at least one year.
